After an open-call tryout is held at Palace Station in Las Vegas, 16 of the best female flyweights from around the world are chosen to compete for the first-ever UFC women's flyweight belt. Upon arrival at the UFC Training Center in Las Vegas, the fighters are greeted by their coaches for the season, former UFC world champion Eddie Alvarez and UFC rising star Justin Gaethje. UFC President Dana White welcome the fighters, introduces the coaches, and explains that this season the winner of the tournament will become the first ever UFC women's flyweight world champion. As the coaches pick their teams the fighters' seeds are revealed, and the fight matchups are revealed. Then, the first preliminary fight is announced. After the fighters move into the TUF house, team training begins leading up to the first preliminary fight of the season.

With the first preliminary fight in the books, Justin Gaethje and a few assistant coaches stop by the house for a night of fun and games. The girls bond with one another and take the opportunity to relax. Justin attempts to talk strategy with an international fighter but finds it quite hard to communicate with her, even with the help of a translator. Meanwhile, the Team Alvarez girls have their own fun in the backyard. One of the fighters has a very peculiar way of going about her weight cut, which turns out to be highly entertaining to the others. The next day, at practice, the fighters see the tournament bracket for the first time and quickly realize they are likely to face a teammate as early as the quarterfinals. Coach Alvarez is upset and tries to keep the team spirit alive after one of his girls asks him to prep her for a potential fight against a teammate. As the training session gets underway, Alvarez focuses his attention on his next flyweight, who is confident that she can dominate the upcoming fight. Tragedy strikes at the Team Gaethje practice as one of his fighters gets injured while doing drills on the mat. She ends up in the ER and receives an uncertain prognosis. Later that night at the house, the injured fighter receives a visit from former UFC champion Miesha Tate, who encourages her to stay positive. As the fight approaches, Justin prepares his next competitor and seems to be extremely confident in her stand up abilities. The day of the fight comes and the two flyweights put on an entertaining show. The second quarterfinalist claims her spot and the coaches announce the next matchup.

Meanwhile, Eddie's next fighter is excited to step back into the octagon after a prolonged absence from the sport. At the Team Gaethje training, Justin seems to be a bit concerned that his next young fighter may rely too heavily on her jiu-jitsu skills. Despite being far less experienced than her upcoming opponent, the fighter believes that she is ready to step up to the challenge. Back at the house, one of the more reserved competitors wakes up feeling very discouraged about the competition. She reveals that she's had an old injury flare up in practice and hopes that it won't affect her going into her first fight. The fighter heads over to the Performance Institute where she receives cupping therapy to ease the pain. She leaves feeling hopeful but still worrying about whether or not she will be able to perform to the best of her abilities. Through home video footage, the upcoming fighters reveal more about their home life, their gyms and how they got started in MMA. The day before the fight, both girls make weight and feel confident going into the bout. The next day, an exciting fight determines the 6th quarterfinalist and the coaches pick the 7th preliminary fight.

The time has come for the coaches challenge and both teams meet up with Dana White at a local pool to watch Eddie and Justin go head to head in an intense race. Eddie and Justin give it their all in the hopes of winning $10,000 for themselves and $1,500 for each of their fighters. As fight day approaches, Team Alvarez's fighter opens up about her life back home with her son. The weigh-in ends with an intense stare down between the two flyweights. The next day, the fight determines who moves on to the next round and the quarterfinals are announced.

At her opponent's team practice, the coaches seem confident that their fighter needs to use her conditioning and take the fight to the later rounds in order to win. Even though she is the favorite on paper, they take nothing for granted and prepare for a tough battle. In the days leading up to the fight, one of the flyweights decides to go to the UFC Performance Institute in order to get help with her weight cut. They assess her health and advise her on the best course of action. The fighter spends the day at the institute under supervision and safely achieves her goal. On fight day, the two flyweights deliver a great show. The winner moves on to the live finale and faces off against her opponent. Finally, Coaches Eddie Alvarez and Justin Gaethje square off in anticipation of their fight at UFC 218.
